The word 'in' is a preposition. A preposition is, in a sense, a connector. When you use the word 'in' in the sentence, you should notice that it smoothly connects the other words around it. If the word at isn't working as a connector, then it will most likely be working as an adverb.

"Although he studied hard, he couldn't succeed in getting a high score in his test."

This option makes the sentence grammatically correct and coherent. It conveys the idea that despite studying hard, the person was not able to achieve a high score on the test.

This would suggest he couldn't succeed "on" the test itself, which doesn't quite fit the context. 

This would imply he couldn't succeed "at" the overall goal of getting a high score, rather than the process of achieving it.

This doesn't properly complete the sentence, as "to getting a high score" doesn't convey the right meaning.
